Assessment of Angiogenesis Promotors and Cytokines in Intraocular Fluid of Patients with Proliferative Diabetic Retinopathy

Abstract:

Purpose:
To determine the concentrations of Vascular Endothelial Growth Factor (VEGF), Epidermal Growth Factor (EGF), Interleukin 6 (IL 6), Interleukin 8 (IL 8), Interferon Gamma- Induced Protein 10kDa (IP 10), Monocyte Chemotactic Protein-1 (MPC1), Platelet-Derivered Growth Factor-AA (PDGFAA), Brain - Derivered Neurotrophic Factor (BDNF), Ciliary Neurotrophic Factor (CNTF) and Transforming Growth Factor Beta (TGFb) in intraocular fluid of patients with complications of proliferative diabetic retinopathy (PDR).

Patient cohort:
50 eyes of 50 patients with complications of PDR and 50 eyes of 50 patients before cataract surgery without any ocular pathology (control group).

Methods:
Intraocular fluid samples were aspirated from anterior chamber of 50 eyes of patients with PDR before surgical or anti-VEGF treatment. Concentrations of angiogenesis promotors and cytokines were compared with levels measured in control group. Angiogenesis promotors levels were assessed by xMAP multiplex analysis. Pair sign test was used for statistic analysis.

Results:
A significantly increased concentrations of VEGF, IL6, IL8 and PDGFAA were observed in PDR patients in comparison with control group. Statistically meaningful higher concentration of antiangiogenic IP 10 was measured in PDR patients in comparison with the control group. Assessed concentrations of EGF, BDNF, CNTF, MPC 1 and TGFb were not significantly differrent in observed groups.

Conclusion:
Concentrations of VEGF and other angiogenic markers were significantly higher in patients with PDR. Also significantly increased level of antiangiogenic IP 10 was observed in PDR patients.
